ANX-201 (thiophosphonoformate)
ANX-201 (thiophosphonoformate) is a reverse transcriptase inhibitor with a novel mechanism of action and unique resistance profile. ANX-201 has been shown in vitro to re-sensitize nucleotide reverse transcriptase inhibitor (NRTI)-resistant virus. ANX-201 is a pyrophosphate analog that binds in the active site of viral polymerases, including reverse transcriptase, inhibiting viral DNA or RNA chain elongation and therefore viral replication. HIV replicates rapidly and can readily mutate to eventually evade inhibitor drugs and drug cocktails. Therefore, new drugs that target novel areas of the virus or overcome drug resistance are needed.The Company plans to file an IND with the FDA in 2007 and to initiate a Phase 1/2 clinical trial of ANX-201 in HIV-infected patients who have resistance to NRTIs. ANX-201 is a broad spectrum antiviral and has shown activity against HIV-1, HIV-2, influenza A and herpes viruses. The Company is continuing to evaluate the broad antiviral properties of ANX-201 in preclinical studies. |
